چکیده

Simple, automated methods are required for rapid detection of wastewater contamination in urban recreational water. The activity of the enzyme β-D-galactosidase (GAL) can rapidly (<2 h) be measured by field instruments, or a fully automated instrument, and was evaluated as a potential surrogate parameter for estimating the level of fecal contamination in urban waters. The GAL-activity in rivers, affected by combined sewer overflows, increased significantly during heavy rainfall, and the increase in GAL-activity correlated well with the increase in fecal indicator bacteria. The GAL activity in human feces (n = 14) was high (mean activity 7ˆ 107 ppb MU/hour) and stable (1 LOG10 variation), while the numbers of Escherichia coli and intestinal enterococci varied by >5 LOG10. Furthermore, the GAL-activity per gram feces from birds, sheep and cattle was 2–3 LOG10 lower than the activity from human feces, indicating that high GAL-activity in water may reflect human fecal pollution more than the total fecal pollution. The rapid method can only be used to quantify high levels of human fecal pollution, corresponding to about 0.1 mg human feces/liter (or 103 E. coli/100 mL), since below this limit GAL-activity from non-fecal environmental sources may interfere.

A sensitive and robust method for automated on-line monitoring of enzymatic activities in water and water resources.

The realisation of a novel concept for automated on-line monitoring of enzymatic activities in water was successfully demonstrated by long-term field testing at two remote Austrian ground water resources. The β-D-glucuronidase (GLUC) activity was selected as a representative enzymatic model parameter for the on-line determination. But the device can be adapted for any enzymatic reaction with di...

Bioaffinity Based Immobilization of Almond (Amygdalus communis) b-galactosidase on Con A-layered Calcium Alginate-cellulose Beads: Its Application in Lactose Hydrolysis in Batch and Continuous Mode

In this study, immobilization of partially purified almond (Amygdalus communis) β-galactosidase on Con A layered calcium alginate-cellulose beads was investigated. Immobilized β-galactosidase retained 72% of theinitial activity after crosslinking by glutaraldehyde. Both soluble and immobilized enzyme exhibited the samepH and temperature optima at pH 5.5 and 50ºC, respectively. Howev...

Evaluation of Avian Waste and Bird Counts as Predicators of Escherichia coli Contamination at Door County, Wisconsin Beaches

Microbial source tracking (MST) has become a focus of some recreational beach monitoring programs. Suspected sources of contamination include human sewage, agricultural runoff, and feces from wildlife and domestic animals, depending on beach location. Waterfowl have been suggested as a primary source of fecal contamination at many beaches, but techniques to “prove” contaminating microbes are of...

Monitoring of fecal pollution in coastal waters by use of rapid enzymatic techniques.

Enzyme assays for 4-methylumbelliferyl-beta-D-galactopyranosidase and 4-methylumbelliferyl-beta-D-glucuronidase activities were used for rapid detection (25 min) of fecal water pollution and to determine the impact of sewage discharge in coastal waters. Two coastal areas were investigated: (i) an estuary characterized by a high degree of contamination downstream of a discharge from a sewage tre...

New Approaches for Enhanced Detection of Enteroviruses from Hawaiian Environmental Waters

Health risks associated with sewage-contaminated recreational waters are of important public health concern. Reliable water monitoring systems are therefore crucial. Current recreational water quality criteria rely predominantly on the enumeration of bacterial indicators, while potentially dangerous viral pathogens often remain undetected. Human enteric viruses have been proposed as alternative...
